What is Inclisiran?

Inclisiran is a relatively new injectable medication used to treat high low-density lipoprotein (LDL) cholesterol levels. You might have heard this referred to as ‘bad’ cholesterol.

Inclisiran belongs to a class of drugs called PCSK9 inhibitors. It works by blocking a type of protein made in your liver that encourages your LDL cholesterol levels to build up. It can be used alone or in combination with other treatments. 

You may be able to use Inclisiran if you can’t get your LDL cholesterol to target levels with other medications, including statins, and have existing cardiovascular disease (heart disease, strokes, or peripheral arterial disease).

Clinical trial evidence shows that Inclisiran can halve high LDL cholesterol levels, with reductions usually seen after 90 days of treatment.

Inclisiran is administered by a cardiac nurse via an injection, usually in your abdomen, upper arm, or thigh. When you first start the drug, you’ll need to have a second dose 3 months after your first. After that, your injections are given every 6 months.

The injection is quick, so you’ll only need to attend our outpatient clinic for a short appointment. As with any injection, you may momentarily feel a sharp scratch. The area where you were injected may be slightly red and feel a little sore for a day or so afterwards. You’ll be able to go home and resume your normal daily activities shortly afterwards.

Inclisiran is considered to be a safe drug, with very few risks. As with any medication, it has some side effects, but these are rare. Side effects may include:

  • joint pain

  • painful urination

  • a cough, wheezing, or breathlessness

  • allergic reaction

  • pain, swelling, and redness at the injection site

Having it administered correctly by one of our cardiac nurses lowers the risks even more. 

Inclisiran is not suitable if you’re pregnant or breastfeeding.

Before your appointment to receive your Inclisiran injection, you’ll have a blood test to confirm the starting levels of cholesterol in your bloodstream. You’ll also have blood tests to check your liver, kidney, and thyroid function, and your diabetes status.

You may also be asked to stop taking certain prescribed medications, over-the-counter medications, or supplements before starting Inclisiran.

Wearing loose clothing to your appointment can make it easier for your cardiac nurse to administer your injection.

You’ll have a follow-up appointment 3 months after your first injection of Inclisiran. At this appointment, you’ll have another blood test to assess your cholesterol levels. If the results show the treatment is working for you, you’ll be able to continue your treatment with a second dose of Inclisiran. After that, you’ll have further injections every 6 months.

Frequently asked questions

Research hasn’t confirmed that Inclisiran is safe and effective for children. So, the medication is available for adults, with no upper age limit, currently.

So far, no clinical trials have found that the medication leads to weight gain.

High cholesterol is usually caused by lifestyle factors such as poor diet (not getting enough nutrients and/or eating too much saturated fat) and limited exercise (not doing enough physical activity). There is also a genetic aspect to having high cholesterol.
